目的:探究腹部手术后焦虑与肠道菌群紊乱之间的关系。 方法:将16只C57BL/6J小鼠随机分为实验组(n = 8)和对照组(n = 8),实验组小鼠行腹部切开,对照组小鼠无任何处理。术后第7天收取小鼠粪便,术后第15天收取小鼠肝脏,肠道等组织,期间记录两组小鼠的体质量,自主活动、认知及焦虑抑郁情况。分析肠道菌群差异,并与术后焦虑抑郁行为学指标进行相关性分析。 结果:术后两组小鼠自主活动和认知能力无显著差异;实验组小鼠焦虑抑郁样行为明显增多(P < 0.05) ,且伴有肠粘膜低度炎症反应。两组小鼠肠道菌群存在显著差异,实验组小鼠粪便中Akkermansia细菌丰度明显降低(P < 0.05),Enterorhabdus细菌的相对丰度显著增加(P < 0.05);相关性分析显示,Akkermansia菌丰度和焦虑行为存在显著负相关(R = 0.52,P = 0.045)。 结论:小鼠腹部切开术后出现焦虑样行为可能与肠道中Akkermansia菌减少相关。
Abstract:
Objective: This study was designed to explore the relationship between postoperative anxiety and gut microflora disorder. Methods: Sixteen C57BL/6J mice were randomly divided into experimental groups (n = 8) and control groups (n = 8). The experimental group underwent abdominal incision and the control group did not receive any treatment. Feces were collected on Day 7 after the surgery, liver and intestine tissues were collected on Day 15 after the surgery. Body weight, autonomous activity, cognition, anxiety and depression in both groups were recorded. Gut microbiota was analyzed, and the correlation between it and anxiety-like behavior after surgery was analyzed . Results: There were no significant differences in autonomic activity and cognitive ability between the two groups. The experimental group mice showed significantly increased anxiety-depressive behavior (P < 0.05) with low inflammation of intestinal mucosa. In the experimental group, the abundance of Akkermansia in the feces decreased and the relative abundance of Enterorhabdus increased significantly (P < 0.05). The correlation analysis showed a significant correlation between Akkermansia abundance was negatively correlated with anxiety-like behavior (R = 0.52, P = 0.045). Conclusion: Anxiety-like behavior after abdominal incision be related to the decrease of Akkermansia in intestine.
