摘要:
目的:初步研究特殊配方肠内营养(EEN)联合普拉梭菌上清(FPS)对2,4,6三硝基苯磺酸(TNBS)诱导的大鼠结肠炎疗效。 方法:40只SD大鼠随机分为5组,每组8只:健康对照组、TNBS诱导结肠炎模型组、不同浓度EEN (12.5%和100%)组及100% EEN+FPS组。造模成功后第1天起至第4天,各组大鼠自由饮食,给药组灌胃给予相应剂量的 EEN 和 FPS。第 5 天处死大鼠。测量结肠长度,计算结肠重量与长度比,行结肠组织病理分析;ELISA法检测血浆及结肠组织IL-1β、IL-6、TNF-α和IL-17水平;结肠组织MPO及iNOS活性检测。组间比较采用单因素方差分析。 结果:模型组较对照组体质量下降,结肠长度缩短,结肠重量与长度比、血清、结肠组织中IL-1β、IL-6、TNF-α和IL-17的表达及结肠组织中MPO和iNOS活性均显著增加。予不同浓度EEN,大鼠体质量逐渐增加,结肠长度逐渐恢复至近正常水平,结肠重量/长度、血清和结肠组织中炎症因子水平以及结肠组织中MPO和iNOS活性均逐渐下降;100% EEN基础上联合+FPS组能更加显著降低结肠重量/长度(P < 0.01),血清和结肠组织中炎症因子的表达及MPO和iNOS活性有显著差异(P < 0.05)。 结论:EEN能显著抑制SD大鼠结肠炎症反应,随着 EEN浓度增加疗效也逐渐增加。100% EEN+FPS较单独100% EEN具有更加显著的结肠炎炎症抑制作用。
Abstract:
Objective: To study the effect of exclusive enteral nutrition (EEN) combined with Faecalibacterium prausnitzii supernatant (FPS) on trinitro-benzen-sulfonic acid (TNBS) induced colitis in rats. Method: 40 SD rats were randomly divided into 5 groups with 8 rats in each group: control group, trinitro-benzen-sulfonic acid (TNBS) group, variable EEN (12.5%and 100%) groups and 100% EEN+ FPS group. Rats in each group were fed ad libitum for 4 days, and rats in EEN (12.5%and 100%) groups and 100% EEN+ FPS group were administered corresponding doses of EEN and FPS by gavage. The rats were sacrificed on the 5th day. Then, the length of colon was measured, the weight/ length of colon was calculated, and a histopathological analysis of the colon was performed. The levels of IL-1β, IL-6, TNF-α and IL-17 in plasma and colon tissues were measured by ELISA. The activities of MPO and iNOS in colon tissues were detected. One way ANOVA was used for the comparison between groups. Results: In the TNBS group, the weight of rats was decreased, the colon was shortened, the weight/ length of colon increased, the expression of IL-1β, IL-6, TNF-α and IL-17 in serum and colon tissues and the activities of MPO and iNOS in colon tissues increased significantly compared to the control group. With the increase of EEN concentration, the body weight increased significantly, the length of colon gradually returned to nearly normal level, and the weight/ length of colon decreased significantly, the level of these inflammatory factors in serum and colon tissue and the activity of MPO and iNOS in colon tissue decreased gradually. Also, compared with the group of 100% EEN alone, 100% EEN+ FPS significantly reduced the weight/ length of colon (P < 0.01), and the expression of inflammatory factors in serum and colon tissue , the activities of MPO and iNOS in colon tissues decreased significantly (P < 0.05). Conclusion: EEN can significantly inhibit the colon inflammatory response , and the effect gradually increases with the increase of EEN concentration. Compared with 100% EEN, 100% EEN + FPS had a more significant inhibitory effect on colitis.
